A General Requirements:
1 Magnet Compatibility: The probe must be fully compatible with a 700 MHz Standard Bore NMR S3 magnet system.
2 Console compatibility: The probe must be fully compatible with AVANCE III console and should be forward compatible with newer consoles (such as AVANCE III HD, NEO, etc.).
3 Shim stack compatibility: The probe must be fully compatible with BOSS 3 shim stack (36 orthogonal shim gradient) and should be compatible with newer versions of shim systems.
4 Probe Type: Four-channel inverse probe, optimized for solution-state NMR applications, including 31P as one of the channels.
B Pulse and detection channels:
1 Supported Nuclei: The probe must support excitation and detection of 1H, 13C, 15N, and 31P nuclei.
2 Lock Channel: A dedicated 2H (deuterium) lock channel is required for field frequency stabilization. The channel should have the capability to perform 2H decoupling during acquisition.
3 Pulse widths should be lesser than the following for respective channels:
3.1 1H pulse width ≤ 10 μs
3.2 13C pulse width ≤ 12 μs
3.3 15N pulse width ≤ 38 μs
3.4 31P pulse width ≤ 28 μs
3.5 2H pulse width ≤ 100 μs
C Sample Compatibility
1 Tubes Supported: The probe should accommodate both 5 mm and 3 mm standard NMR tubes, with appropriate adaptors or inserts as needed.
D Gradient and Experiment Capability
1 Z-axis gradient capability: Integrated z-axis gradient coil for high-resolution gradient-based experiments. The gradient strength should be 45 G/cm or higher.
2 Experiment Types: Suitable for inverse 1D, 2D, 3D, and such multidimensional NMR experiments, including but not limited to HSQC, HMQC, HMBC, NOESY, and TOCSY.
E Tuning, Matching, and Shimming
1 Tuning and Matching: Automatic and manual tuning and matching of all observed and decoupled nuclei, with user override for manual adjustments.
2 Shimming: Support for both automatic and manual shimming processes to optimize field homogeneity. The probe must be able to perform gradient based shimming.
F Temperature Control
1 Variable Temperature Range: The probe must support variable temperature (VT) experiments across a range from -150°C to +150°C.
2 Temperature Stability: Ensure precise temperature control and stability within ±0.1°C across the specified range.
G Software and Automation
1 Control Software: Full integration and remote control via TopSpin software (version 3.5 or higher).
2 Automation Compatibility: Must be compatible with ICON-NMR for automated experiment setup, execution, and data handling.
3 Autosampler compatibility: Must be compatible with SAMPLECASE autosampler system.
4 Operational Modes: Support both manual and automated operation modes for flexible user control.
H Sensitivity and line shapes
1 1H sensitivity: The probe must provide a 1H sensitivity of at least 1400 (as defined by the sample-to-noise ratio for a standard 1% ethylbenzene in CDCl3 sample, using a 5 mm tube at 25°C, 1 scan).
