BamHI Size: 50000 units BamHI is a Type II restriction endonuclease that recognizes the palindromic DNA sequence 5′-GGATCC-3′ and cleaves within this recognition site. It cuts between the first two guanine residues (G^GATCC), generating cohesive 5′ overhangs (sticky ends). This property makes BamHI widely used in molecular cloning for precise DNA fragment insertion and plasmid construction. The enzyme requires Mg²⁺ as a cofactor and functions optimally under recommended buffer conditions at 37 °C. BamHI can be heat-inactivated (commonly at 65 °C for 20 minutes, depending on the supplier) and is typically supplied in glycerol for storage at −20 °C. It is also important to maintain optimal reaction conditions to avoid star activity, which can lead to non-specific cleavage.
