To analyse particular microorganisms, prepare the solution using weighing certified chemicals. For serial dilution, required test tubes, petriplates and media which are to be autoclaved is prepared.
Serial dilution is done in laminar air flow by using prepared test tubes and petriplates. Place the media in petriplates and inoculate it. seal with cling wrap.

Mix the cowdung with water twice (after 15 to 12 days) and spray on agriculture waste in the pit.

To prepare the VAM culture in pots (12" pots), strelize it and fill it with the mixture of sand, soil and manure in 1:1:1 ratio.

Allow to grow Ragi/Sorghum plant for 75 days (2 ½ month). Cut the plants from base mix with soil along with roots. The prepared VAM culture is used to prepare the VAM biofertilizer.
The beds used for VAM preparation, are cleaned and spread with black colour polythene sheet. Prepare the media of bed with mixture of soil, sand, vermicullite and farm yard manure. Prepared beds (25' X 4' X 1' measurement) are covered with black colour polythene sheet for a week.
After 1 week, prepared VAM culture is mixed with beds with specified quantity. Ragi/Sorghum/any other seeds are sown into the beds.
Water the crop for whole growing period in pots. Irrigate the sown beds for 8 weeks (56 days) for whole crop life cycle.
Weed the bed 4 times (for whole crop life cycle).
After 8 to 10 weeks (56 to 70days), harvest the crop and incorporate into the beds and leave it for 7 days. Later VAM roots are powdered and shade dried, packed into 5/10/20 Kg bags and sealed.
To prepare biomix, the vermicompost is sieved and stored. The stored vermicompost manure is added with specified quantity of Arka Microbial Consortium Biofertilizers.
Mixed biomix is weighed and filled into 5kg / 10kg poly sack bags. Seal the weighed bags and stack them in a room with cool temperature.
Pesticide residue analysis :
Clean the samples and rinse with water. Take the specified amount of the samples and chop into small pieces. The chopped samples are grinded in a grinder with distilled water, then transferred to a jar bottle, labelled and stored.
Preparation of muffle furnace for activation of certain chemicals. Before analysing in LCMS/MS instrument, prepare the mobile phase (mixing the specified chemicals) and sonificate in the sonicator instrument.
Before performing the analysis in the GCMS/MS, Baseline correction is performed by stabilizing the instrument. After switching on the instrument, generate the vacuum and bring the instrument to a stabilization stage.
Weigh the Certified Reference Materials precisely for preparation of primary standards. Then transfer into volumetric flasks to prepare the working standards as per SoP and store it in the Deep Freezer.
Prepare the instrument by calibrating for analysis. Weigh the stored sample accurately. Add required solvent and distilled water to the weighed sample. To that sample add required chemicals and mix well.
Clean the homogenizer in distilled water and homogenize the blended sample. Centrifuge the homogenized sample and transfer the supernatant into vortex and re-centrifugate and filter.
For analysis of pesticide residues in LCMS, reconstitute the solvent in nitrogen concentrator and tranfer the sample into 1 ml vials

Description of works
Collection of soil, water, leaf and organic manure samples from technical personnel.
